The short version of ACTH(4-10) fits in a sentence. The long version — which is the one that helps — is below.

Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. Its design combines the ACTH(4-7) core fragment with a C-terminal Pro-Gly-Pro extension, a modification intended to improve stability and prolong activity. The molecule is hydrophilic, carries no lipid chains or glycosylation, and has a theoretical mass just over 810 daltons in its free form. All seven residues are proteinogenic amino acids, so no non-natural building blocks appear in the backbone. A free N-terminal methionine and C-terminal proline define the unmodified parent peptide.
The compound was developed during the 1980s at the Institute of Molecular Genetics in Moscow as part of research on fragments of adrenocorticotropic hormone. Early work examined short ACTH-derived sequences that retained neurotrophic effects while lacking the endocrine activity of the full hormone. Semax entered clinical use in Russia during the 1990s, where it received registration for several neurological indications. Outside that region it remained primarily a laboratory research material rather than an approved therapeutic. English-language literature on it grew more slowly and frequently cited the original Russian studies.
The peptide is prone to several degradation pathways. Oxidation of the methionine residue produces a sulfoxide that elutes close to the parent peak in many chromatographic systems. Hydrolysis of peptide bonds and deamidation of susceptible residues in related sequences also reduce purity over time. Lyophilised material kept dry at minus twenty degrees Celsius and shielded from light is the most stable form commonly described in laboratory practice.
Material sold for laboratory use varies widely in stated purity and documentation. A certificate of analysis should list the analytical method, the column and detector used, and the observed purity value. Independent verification by an outside laboratory is the practical way to confirm identity when documentation is absent or internally inconsistent. Regulatory status differs by country, and a product legal in one jurisdiction may be unapproved or controlled in another.
Identity and purity of semax are established with reversed-phase high-performance liquid chromatography coupled to ultraviolet detection, usually at 214 nanometres. Mass spectrometry, most often electrospray ionisation in positive mode, confirms the molecular mass and reveals truncated sequences. Amino acid analysis and peptide mapping after enzymatic digestion provide additional structural confirmation. Laboratories typically report purity as the percentage area of the main peak, a figure that does not capture isomeric or oxidised variants unless the method resolves them.

Reported pharmacological work centers on neurotrophic signaling, including changes in BDNF and NGF expression in hippocampal tissue in animal models. Human data come largely from studies conducted in Russia, and how well those results generalize to other populations remains an open question. Regulatory status differs sharply by jurisdiction: Semax is a registered prescription medicine in Russia, while it holds no approved marketing status in the United States or the European Union. Outside such jurisdictions it is generally handled as a research chemical, which affects both documentation and quality expectations.
Semax is a synthetic heptapeptide whose sequence is Met-Glu-His-Phe-Pro-Gly-Pro. It was developed as a fragment analog of adrenocorticotropic hormone, modeled specifically on the ACTH(4-10) region. The first four residues reproduce that fragment, while a Pro-Gly-Pro tripeptide is appended at the C-terminus. Work on the compound originated in Russia, where it entered clinical use as an intranasal preparation. Its sequence places it among short regulatory peptides studied for effects on the central nervous system rather than on the adrenal axis.
The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.
Russian regulatory authorities approved the peptide for nasal administration, and it remains listed in the national pharmacopoeia under several trade names. Documented indications include acute ischemic stroke, transient ischemic attacks, traumatic brain injury, and certain ophthalmological and neurological conditions. Physicians also prescribe it for cognitive complaints in older patients, although the evidence base for that use is thinner. Outside Russia and a few neighboring states, the substance is not an approved medicine and is sold instead as a research chemical.
Later generations of the molecule include an N-acetylated form and an amidated form, both marketed online alongside the parent peptide. These variants differ in terminal chemistry and stability, and they are frequently discussed in the same breath even though they have not been compared in controlled trials. Supply outside formal healthcare systems comes largely from laboratories that synthesize peptides to order. Purity and identity of these materials vary widely, and no single body oversees the international trade.
Semax is a synthetic heptapeptide developed in the Soviet Union during the 1980s by researchers at the Institute of Molecular Genetics in Moscow. It was designed as a truncated analog of adrenocorticotropic hormone, retaining only the fragment spanning residues four through ten. Investigators sought a peptide that would preserve the cognitive effects associated with ACTH while eliminating the hormonal stimulation of the adrenal cortex. The compound entered clinical use in Russia during the following decade.
Identity and purity are commonly assessed by reversed-phase high-performance liquid chromatography, with ultraviolet detection near 214 nanometers for the peptide backbone. Mass spectrometry, either electrospray or matrix-assisted laser desorption, confirms molecular mass and detects truncation or modification products. Amino acid analysis can verify composition. Because the sequence contains no strongly absorbing aromatic residue apart from phenylalanine, detection wavelengths are chosen carefully. Purity values above 95 percent are typical for research-grade material.
Quality documentation for research-grade peptide usually includes a certificate of analysis stating purity, identity, and residual counterion content. Batch-to-batch variation in purity and salt form is a recognized issue, and comparisons across suppliers require attention to the exact counterion, for example acetate versus trifluoroacetate. Sequence verification by tandem mass spectrometry provides stronger evidence than a single mass measurement. For any study reporting biological results, the analytical method and the measured purity should be stated explicitly. Independent verification of supplier claims is considered good practice.
===== Human T-lymphotropic virus (types I and II) ===== Human T-Lymphotrophic Virus (HTLV) is able to be passed through breastmilk from mother to child. The worldwide rate of transmission through breastmilk is estimated to be 3.9–27%, and this risk is increased by high maternal viral load and prolonged periods of breastfeeding. Current data demonstrates that while breastfeeding for less than six months does not, independently, increase risk of HTLV-1 transmission, not breastfeeding during that time does decrease risk of transmission. As such, CDC recommends against breastfeeding when mothers have HTLV Types I or II. Recognizing the importance of breastfeeding in more resource-poor areas of the world, the World Health Organization recommends shortening the duration of breastfeeding, or avoiding breastfeeding where possible.
== History == GABA was first synthesized in 1883; it was first known only as a plant and microbe metabolic product. In 1950, Washington University School of Medicine researchers Eugene Roberts and Sam Frankel used newly developed techniques of chromatography to analyze protein-free extracts of mammalian brain. They discovered that GABA is metabolized from glutamic acid and accumulates in the mammalian central nervous system. There was not much further research into the substance until 1957; Canadian researchers identified GABA as the mysterious component (termed Factor I by its discoverers in 1954) of brain and spinal cord extracts which inhibited crayfish neurons. In 1959, it was shown that, at an inhibitory synapse on crayfish muscle fibers, GABA acts through stimulation of the inhibitory nerve. Both inhibition by nerve stimulation and by applied GABA are blocked by picrotoxin.
== History == After the failure of the Embargo Act of 1807, the federal government of the United States took little interest in imposing embargoes and economic sanctions against foreign countries until the 20th century. United States trade policy was entirely a matter of economic policy. After World War I, interest revived. President Woodrow Wilson promoted such sanctions as a method for the League of Nations to enforce peace. However, he failed to bring the United States into the League, and the U.S. did not join the 1935 League sanctions against Italy. According to communications studies academic Stuart H. Davis and political scientist Immanuel Ness, trends in whether the United States has unilaterally or multilaterally imposed sanctions have changed over time. During the Cold War, the United States led unilateral sanctions against Cuba, China, and North Korea. Following the disintegration of the Soviet Union and the end of the Cold War, United States sanctions became increasingly multilateral. During the 1990s, the United States imposed sanctions against countries it viewed as rogue states—such as Zimbabwe, Yugoslavia, and Iraq—in conjunction with multilateral institutions such as the United Nations or the World Trade Organization. Davis and Ness state that in the 2000s, and with increasing frequency in the 2010s, the United States acted less multilaterally as it imposed sanctions against perceived geopolitical competitors such as Russia and China, or countries that were the site of "proxy conflicts" such as Yemen and Syria.

Alcohol has a variety of short-term and long-term adverse effects. Alcohol has both short-term, and long-term effects on the memory, and sleep. It also has reinforcement-related adverse effects, including alcoholism, dependence, and withdrawal. Alcohol use is directly related to considerable morbidity and mortality, for instance due to intoxication and alcohol-related health problems. The World Health Organization advises that there is no safe level of alcohol consumption. Many of the toxic and unpleasant actions of alcohol in the body are mediated by its carcinogenic byproduct acetaldehyde.
19 December – With the UK government signalling its willingness to delay local elections due in May 2026 for a year, the Electoral Commission raises concerns about further delays to elections, describing the process as one that is "damaging public confidence". 21 December – The Electoral Commission finds no "credible evidence of potential offences of electoral law" over Nigel Farage's election expenses. Scottish Labour MSP Pam Duncan-Glancy confirms she will not seek re-election to the Scottish Parliament at the 2026 election because she does not want her friendship with a convicted sex offender to "become a distraction". 22 December – Liberal Democrat leader Sir Ed Davey writes to the Equalities and Human Rights Commission to ask it to investigate government plans to delay local elections, which could affect as many as 10 million voters. The leader of Plymouth City Council confirms that local elections there will go ahead in May, but local elections in Exeter are still uncertain. 23 December – The UK government announces it is watering down its plans for inheritance tax on farmers, and will raise the threshold from £1m to £2.5m. Local Government Secretary Steve Reed writes to councils in England warning them not to adopt a four-day working week. Scotland's Justice Secretary, Angela Constance, is to face an investigation over comments she made about a grooming gang expert to determine whether she broke the ministerial code.
== Connections with Mauritania == In testimony given by Antonio Panzeri in February 2023, he and Francesco Giorgi received €200,000 from Mauritania to improve the image of the country in the EU, as requested by then-president, Mohamed Ould Abdel Aziz in 2018. Aziz left office in 2019, with Mohamed Ould Ghazouani winning the election to become president. Paid €50,000 per annum from 2019, Panzeri helped organise an international conference in Mauritania in June 2022. Mauritania only abolished slavery in 1981 and had been criticised by the EU for their human rights record, yet received millions annually from the EU to host fishing vessels. Marie Arena, as head of the EU subcommittee on human rights, proposed the conference and invited Panzeri, who was no longer an MEP to attend. Panzeri and Giorgi were speakers and Andrea Cozzolino also attended, Arena and Cozzolino meeting with the President of Mauritania, Ghazouani.
== History == In 1976, Gustilo and Anderson refined the early classification system proposed by Veliskasis in 1959. An early study conducted by Gustilo in 1976 showed that primary closures with prophylactic antibiotics of Type I and type II fractures reduced the risk of infection by 84.4%. Meanwhile, early internal fixation and primary closure of the wound in Type III fractures have a greater risk of getting osteomyelitis. However, Type III fractures occur in 60% of all the open fracture cases. Infection of the Type III fractures is observed in 10% to 50% of the time. Therefore, in 1984, Gustilo subclassified Type III fractures into A, B, and C with the aim of guiding the treatment of open fractures, communication and research, and to predict outcomes. Based on the results of the previous studies, Gustilo initially recommended therapeutic irrigation and surgical debridement for all fractures with primary closure for Type I and II fractures; secondary closure without internal fixation for Type III fractures. However, soon after that, he recommended internal fixation devices for Type III fractures.
== Mechanism == At the cellular level, metabolic myopathies lack some kind of enzyme or transport protein that prevents the chemical reactions necessary to create adenosine triphosphate (ATP). ATP is often referred to as the "molecular unit of currency" of intracellular energy transfer. The lack of ATP prevents the muscle cells from being able to function properly. Some people with a metabolic myopathy never develop symptoms due to the body's ability to produce enough ATP through alternative pathways (e.g. the majority of those with AMP-deaminase deficiency are asymptomatic). H2O + ATP → H+ + ADP + Pi + energy → muscle contraction ATP is needed for muscle contraction by two processes:

It is a synthetic seven-amino-acid peptide built from the ACTH(4-7) sequence plus a Pro-Gly-Pro tail. It belongs to the family of short ACTH fragments studied for nervous-system effects. Its backbone contains only standard amino acids.
It contains the 4-7 fragment of adrenocorticotropic hormone extended at the C terminus. That truncation removes the region associated with steroidogenic activity. The design intent was to keep neurotrophic properties while avoiding full hormonal effects.
No natural source of the intact heptapeptide is known. It is produced by solid-phase peptide synthesis. Some related ACTH fragments arise as hormone processing products, but the Pro-Gly-Pro extension is a designed addition rather than a natural sequence.
Reversed-phase high-performance liquid chromatography is the standard method, with detection in the ultraviolet range. Peak area percentage yields a purity figure for the main component. Mass spectrometry is normally run alongside to confirm molecular identity.
