Identity and purity of semax are established with reversed-phase high-performance liquid chromatography coupled to ultraviolet detection, usually at 214 nanometres. Mass spectrometry, most often electrospray ionisation in positive mode, confirms the molecular mass and reveals truncated sequences. Amino acid analysis and peptide mapping after enzymatic digestion provide additional structural confirmation. Laboratories typically report purity as the percentage area of the main peak, a figure that does not capture isomeric or oxidised variants unless the method resolves them.
The peptide is prone to several degradation pathways. Oxidation of the methionine residue produces a sulfoxide that elutes close to the parent peak in many chromatographic systems. Hydrolysis of peptide bonds and deamidation of susceptible residues in related sequences also reduce purity over time. Lyophilised material kept dry at minus twenty degrees Celsius and shielded from light is the most stable form commonly described in laboratory practice.
Semax is a synthetic heptapeptide whose sequence is Met-Glu-His-Phe-Pro-Gly-Pro. It was developed as a fragment analog of adrenocorticotropic hormone, modeled specifically on the ACTH(4-10) region. The first four residues reproduce that fragment, while a Pro-Gly-Pro tripeptide is appended at the C-terminus. Work on the compound originated in Russia, where it entered clinical use as an intranasal preparation. Its sequence places it among short regulatory peptides studied for effects on the central nervous system rather than on the adrenal axis.
The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.

Later generations of the molecule include an N-acetylated form and an amidated form, both marketed online alongside the parent peptide. These variants differ in terminal chemistry and stability, and they are frequently discussed in the same breath even though they have not been compared in controlled trials. Supply outside formal healthcare systems comes largely from laboratories that synthesize peptides to order. Purity and identity of these materials vary widely, and no single body oversees the international trade.
Semax is a synthetic heptapeptide developed in the Soviet Union during the 1980s by researchers at the Institute of Molecular Genetics in Moscow. It was designed as a truncated analog of adrenocorticotropic hormone, retaining only the fragment spanning residues four through ten. Investigators sought a peptide that would preserve the cognitive effects associated with ACTH while eliminating the hormonal stimulation of the adrenal cortex. The compound entered clinical use in Russia during the following decade.
Russian regulatory authorities approved the peptide for nasal administration, and it remains listed in the national pharmacopoeia under several trade names. Documented indications include acute ischemic stroke, transient ischemic attacks, traumatic brain injury, and certain ophthalmological and neurological conditions. Physicians also prescribe it for cognitive complaints in older patients, although the evidence base for that use is thinner. Outside Russia and a few neighboring states, the substance is not an approved medicine and is sold instead as a research chemical.
=== Detection in body fluids === Buprenorphine and norbuprenorphine may be quantified in blood or urine to monitor use or non-medical recreational use, confirm a diagnosis of poisoning, or assist in a medicolegal investigation. A significant overlap of drug concentrations exists in body fluids within the possible spectrum of physiological reactions ranging from asymptomatic to comatose. Therefore, knowing both the route of administration of the drug and the level of tolerance to opioids of the individual is critical when results are interpreted.
== Location == Using radioimmunoassay and immunocytochemistry, endomorphins have been localized within the nervous systems of humans, mice, rats, and monkeys. Both endomorphin tetrapeptides are abundant in the human neocortex. Endomorphin-1 can be found in the hypothalamus and thalamus of the diencephalon, and in the striatum and lateral septum of the telencephalon. In the hindbrain, endomorphin-1-reactive neurons are more abundant than are those immunoreactive for endomorphin-2. Endomorphin-2 is predominantly found in the spinal cord, specifically in presynaptic terminals of afferent neurons in the dorsal horn region. It has been found co-localized with calcitonin and with the pain-conveying neurotransmitter, substance P. Neither endomorphin-1 nor endomorphin-2 has been identified in the amygdala or the hippocampus.
Amphetamine-Dextroamphetamine is known to hamper appetite. === Metal ion binding === Beyond the iron-sulfur proteins, many other metal cofactors in enzymes are bound to the thiolate substituent of cysteinyl residues. Examples include zinc in zinc fingers and alcohol dehydrogenase, copper in the blue copper proteins, iron in cytochrome P450, and nickel in the [NiFe]-hydrogenases. The thiol group also has a high affinity for heavy metals, so that proteins containing cysteine, such as metallothionein, will bind metals such as mercury, lead, and cadmium tightly. Silver ions can also bind tightly to cysteine and cause secondary structure changes in cysteine-containing peptides, e.g. leading to alpha helix formation. This is also the case for zinc fingers in which silver ions can replace zinc and form stable zinc/cysteine clusters with partial alpha helical structure. The extent of alpha helix formation increases with the number of cysteines.
=== Mechanism of action === Phentolamine acts as a reversible dual or non-selective α1- and α2-adrenergic receptor antagonist. Its primary action is vasodilation due to α1-adrenergic receptor blockade. Reportedly, phentolamine may also stimulate β-adrenergic receptors. Non-selective α-blockers can cause a much more pronounced reflex tachycardia than the selective α1 blockers. Like the selective α1 blockers, phentolamine causes a relaxation of systemic vasculature, leading to hypotension. This hypotension is sensed by the baroreceptor reflex, which results in increased sympathetic nerve firing on the heart, releasing norepinephrine. In response, the β1 adrenergic receptors on the heart increase their rate, contractility, and dromotropy, which help to offset the decrease in systemic blood pressure. Unlike the α1 selective blockers, phentolamine also inhibits the α2 receptors, which function predominantly as presynaptic negative feedback for norepinephrine release. By abolishing this negative feedback phentolamine leads to even less regulated norepinephrine release, which results in a more drastic increase in heart rate.
In polyarteritis nodosa, small aneurysms are strung like the beads of a rosary, therefore making this "rosary sign" an important diagnostic feature of the vasculitis. The 1990 ACR criteria were designed for classification purposes only, but their good discriminatory performance, indicated by the initial ACR analysis, suggested their potential usefulness for diagnostic purposes as well. Subsequent studies did not confirm their diagnostic utility, demonstrating a significant dependence of their discriminative abilities on the prevalence of the various vasculitides in the analyzed populations. Recently, an original study, combining the analysis of more than 100 items used to describe patients' characteristics in a large sample of vasculitides with a computer simulation technique designed to test the potential diagnostic utility of the various criteria, proposed a set of eight positively or negatively discriminating items to be used as a screening tool for diagnosis in patients suspected of systemic vasculitis.
Posho or kawunga—called ugali in Kenya, it is usually made from maize but also other starches; regional names include kwon. Ugandan expatriates make posho from cornmeal, masa harina or grits. Kwon is a type of ugali made from millet (called kalo in western Uganda), but in other regions like eastern Uganda they include cassava flour. Groundnuts (peanuts)—groundnuts are a vital staple and groundnut sauce is extremely common in Ugandan cuisine. Groundnuts are eaten plain or mixed with smoked fish, smoked meat or mushrooms, and can also be mixed with greens such as borr. Sim-sim (sesame)—a staple particularly in the north, roasted sesame paste is mixed into a stew of beans or greens and served as a side dish, though sesame paste may also be served as a condiment; a candy is made from roasted sesame seeds with sugar or honey. Matooke—(green banana, not plantain) boiled or steamed (mashed) cooked in or served with a sauce of peanuts, beans, fresh fish or meat Luwombo—a traditional dish from Buganda, in which a stew of chicken, beef, mushrooms or fish is steamed in banana leaves Malewa—a traditional dish from eastern Uganda (Bugisu), made from bamboo shoots Kikomando—a chapati that is cut into pieces and served with fried beans

Reversed-phase high-performance liquid chromatography is the standard method, with detection in the ultraviolet range. Peak area percentage yields a purity figure for the main component. Mass spectrometry is normally run alongside to confirm molecular identity.
Dry lyophilised powder is generally kept at minus twenty degrees Celsius, protected from light and moisture. Repeated freeze-thaw cycles are avoided because they encourage aggregation and moisture uptake. Solutions are less stable than the solid form and degrade faster at room temperature.
A certificate documents what the supplier measured, not what an independent party confirmed. Methods, instruments and acceptance criteria can differ between suppliers. Third-party testing is the usual way to resolve discrepancies.
The sequence is Met-Glu-His-Phe-Pro-Gly-Pro, a seven-residue peptide. The first four residues correspond to the ACTH(4-10) fragment, and Pro-Gly-Pro is added at the C-terminus. N-acetylated versions also appear in the literature and differ in mass.
