heptapeptide raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.

The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.
Reported pharmacological work centers on neurotrophic signaling, including changes in BDNF and NGF expression in hippocampal tissue in animal models. Human data come largely from studies conducted in Russia, and how well those results generalize to other populations remains an open question. Regulatory status differs sharply by jurisdiction: Semax is a registered prescription medicine in Russia, while it holds no approved marketing status in the United States or the European Union. Outside such jurisdictions it is generally handled as a research chemical, which affects both documentation and quality expectations.
Semax is a synthetic heptapeptide whose sequence is Met-Glu-His-Phe-Pro-Gly-Pro. It was developed as a fragment analog of adrenocorticotropic hormone, modeled specifically on the ACTH(4-10) region. The first four residues reproduce that fragment, while a Pro-Gly-Pro tripeptide is appended at the C-terminus. Work on the compound originated in Russia, where it entered clinical use as an intranasal preparation. Its sequence places it among short regulatory peptides studied for effects on the central nervous system rather than on the adrenal axis.

Development of the compound took place in the Soviet Union during the 1980s, largely through peptide research groups affiliated with the Shemyakin-Ovchinnikov Institute and Moscow State University. Investigators screened fragments of adrenocorticotropic hormone for activity on the central nervous system while attempting to separate cognitive effects from hormonal ones. The shortened sequence was selected because it retained behavioral activity in animal models without stimulating corticosteroid release. Early publications described the molecule as an ACTH(4-10) analog.
Published research on this peptide originates mainly from Russian laboratories, and the wider international literature is comparatively thin. Studies have used rodent models of stroke, hypoxia, and memory tasks, with a smaller number of human trials reported. Reported outcomes include changes in attention and memory measures, along with effects on neurotrophic factor expression in some experiments. Small sample sizes, inconsistent dosing protocols, and limited independent replication are widely noted limitations, so the strength of the clinical evidence remains an open question.
Regulatory status varies sharply by country. Semax is registered for medical use in Russia, where it appears in formularies as a nasal solution, and it also holds registration in a small number of neighbouring states. It has no approval from the United States Food and Drug Administration or the European Medicines Agency, and it is not a scheduled controlled substance in most jurisdictions. Elsewhere it circulates mainly as laboratory material, so purity documentation comes from suppliers rather than from a national pharmacopoeia.
Semax is a synthetic peptide created in the Soviet Union during the early 1980s by researchers working in Moscow. It was built from the short adrenocorticotropic hormone fragment known as ACTH(4-10), and the chain was then extended with three additional amino acids. The resulting molecule was named semax and entered clinical use in Russia in 1994. It is generally described as a nootropic and neuroprotective agent rather than as a hormone analogue.
The level of lipophilicity can be significantly modulated by lipidation, and since lipophilicity is detrimental for the absorption, distribution, metabolism, and excretion of drugs, it provides a way of fine tuning peptides for use in therapeutics.
===== Lawsuits ===== In October 2017, The New Yorker published a story on Mortimer Sackler and Purdue Pharma regarding their ties to the production and manipulation of the oxycodone markets. The article links Raymond and Arthur Sackler's business practices with the rise of direct pharmaceutical marketing and eventually to the rise of addiction to oxycodone in the United States. The article implies that the Sackler family bears some responsibility for the opioid epidemic in the United States. In 2019, The New York Times ran a piece confirming that Richard Sackler, the son of Raymond Sackler, told company officials in 2008 to "measure our performance by Rx's by strength, giving higher measures to higher strengths". This was verified with documents tied to a lawsuit – which was filed by the Massachusetts attorney general, Maura Healey – claiming that Purdue Pharma and members of the Sackler family knew that high doses of OxyContin over long periods would increase the risk of serious side effects, including addiction. Despite Purdue Pharma's proposal for a US$12 billion settlement of the lawsuit, the attorneys general of 23 states, including Massachusetts, rejected the settlement offer in September 2019.
Trilaciclib, sold under the brand name Cosela, is a medication used to reduce the frequency of chemotherapy-induced bone marrow suppression. The most common side effects include fatigue; low levels of calcium, potassium and phosphate; increased levels of an enzyme called aspartate aminotransferase; headache; and infection in the lungs (pneumonia). Trilaciclib may help protect bone marrow cells from damage caused by chemotherapy by inhibiting cyclin-dependent kinase 4/6, a type of enzyme. Trilaciclib is the first therapy in its class and was approved for medical use in the United States in February 2021. The U.S. Food and Drug Administration considers it to be a first-in-class medication. Chemotherapy drugs are designed to kill cancer cells but can damage normal tissues as well. The bone marrow is particularly susceptible to chemotherapy damage. The bone marrow makes red blood cells, white blood cells, and platelets (small fragments in the blood) that transport oxygen, fight infection, and stop bleeding. When damaged, the bone marrow produces fewer of these cells, leading to fatigue, increased risk of infection, and bleeding, among other problems. Trilaciclib may help protect the normal bone marrow cells from the harmful effects of chemotherapy.
Serotonylation has been reported in various cell types and tissues, including both serotonergic and dopaminergic neurons, enterochromaffin cells, cancer-associated fibroblasts, pancreatic 𝛽-cells, CD8+ T cells, pulmonary endothelial cells, platelets, neutrophils, mammary epithelial cells, vascular smooth muscle, and cells of the intestines. Serotonylation is known to influence both tumorigenesis and cancer metastasis, and has been implicated in several types of cancer, including colorectal cancer, neuroendocrine prostate cancer, pancreatic cancer, hepatocellular carcinoma, and ependymomas (brain cancer).
Pneumatic tubes (or capsule pipelines, also known as pneumatic tube transport or PTT or PTS or pneumatic tube systems ) are systems that propel cylindrical containers/carriers through networks of tubes by compressed air or by partial vacuum. They are used for transporting solid objects, as opposed to conventional pipelines which transport fluids. In the late 19th and early 20th centuries pneumatic tube networks were most often found in offices that needed to transport small, urgent packages such as mail, other paperwork, or money over relatively short distances; with most systems confined to a single building or at most an area within a city. The largest installations became quite complex in their time, but have mostly been superseded by digitisation in the information age. Some systems have been further developed in the 21st century in places such as hospitals, to send blood samples and similar time-sensitive packages to clinical laboratories for analysis. A small number of pneumatic transportation systems were built for larger cargo, to compete with train and subway systems. However these systems never gained popularity.

In chemistry, aminolysis (/am·i·nol·y·sis/) is any chemical reaction in which a molecule is lysed (split into two parts) by reacting with ammonia (NH3) or an amine. The case where the reaction involves ammonia may be more specifically referred to as ammonolysis.
== Imaging techniques == Certain issues impact the choice of imaging technique for visualizing soft tissue extracellular matrix (ECM) components. The accuracy of the image analysis relies on the properties and the quality of the raw data and, therefore, the choice of the imaging technique must be based upon issues such as:
Hemostasis is a natural mechanism for minimizing blood loss due to vessel damage while ensuring adequate blood circulation in the organism under Physiologically normal conditions. Platelet adhesion to the exposed collagen due to the action of integrin α2β1 is one of the mechanisms of primary hemostasis. In the state of physiological health, collagen is present below the endothelium and is not accessible to platelets circulating in the blood stream. The damage to the walls of the vessel makes collagen accessible for the platelets and allows them to adhere to collagen fibers due to the activity of platelet collagen receptors, such as α2β1 integrin and GPVI. In contrast to the GPVI functioning as a signaling receptor, α2β1 mainly provides the stabilization of the platelet adhesion to the collagen and facilitates their stable fixation on the damaged area. This process is especially necessary in case of high shear stresses in the arteries due to the negative impact of the blood flow force on platelets' fixation. The enhanced adhesion allows attracting more platelets and forming a stable thrombus that is capable of minimizing blood loss.

== Pharmacokinetics == Endogenous GLP-1 has a plasma half-life of 1.5–2 minutes due to degradation by the ubiquitous enzymes, dipeptidyl peptidase-4 (DPP4) and neutral endopeptidases (NEP). The half-life after intramuscular injection is approximately half an hour, so even administered this way, it has limited use as a therapeutic agent. The metabolically active forms of GLP-1 are the endogenous GLP-1-(7-36)NH2 and the more rare GLP-1-(7-37). The prolonged action of liraglutide is achieved by attaching a fatty acid molecule at one position of the GLP-1-(7-37) molecule, enabling it to both self-associate and bind to albumin within the subcutaneous tissue and bloodstream. The active GLP-1 is then released from albumin at a slow, consistent rate. Albumin binding also results in slower degradation and reduced renal elimination compared to that of GLP-1-(7-37).
==== Thyrotropin-releasing hormone ==== Thyrotropin-releasing hormone (TRH) is a potent stimulator of prolactin release, acting via phospholipase C to mobilise intracellular calcium and activate protein kinase C. In primary hypothyroidism, elevated TRH stimulates both TSH and prolactin, producing hyperprolactinaemia in approximately 20–40% of hypothyroid patients. However, TRH-knockout mice display normal prolactin levels, indicating that TRH is a modulator rather than an obligate releasing factor.
1 August 2024 Southport stabbing The 17-year-old suspect is named as Axel Rudakubana. He is charged with three counts of murder, ten counts of attempted murder, and one count of possession of a bladed article. As a minor, he could not initially be named; but reporting restrictions on his identity are lifted by Liverpool Crown Court. After being charged, Rudakubana is remanded in custody until a plea hearing on 25 October. Keir Starmer holds a crisis meeting with police chiefs and an emergency press conference in the wake of rioting following the murders. Starmer condemns "far-right hatred" responsible for the unrest, and announces the launch of a national violent disorder programme that will see greater cooperation between police forces to tackle violent disorder, as fears grow of a "summer of unrest" fuelled by misinformation. GPs in England vote to take industrial action by working-to-rule over a lack of funding and a decline in care, which could see GP appointments capped at 25 per day. 2 August The BBC reports that Labour has shelved £1.3bn of funding promised by the Conservatives for technology and artificial intelligence (AI) projects, including an exascale supercomputer at Edinburgh University. 2024 Summer Olympics: Team GB secures three gold medals – in rowing, equestrian, and trampoline – bringing their current total to 25 medals: nine golds, eight silvers and eight bronzes.
== Signs and symptoms == In their early stages, soft-tissue sarcomas usually do not cause symptoms. Because soft tissue is relatively elastic, tumors can grow rather large, pushing aside normal tissue, before they are felt or cause any problems. The first noticeable symptom is usually a painless lump or swelling. As the tumor grows, it may cause other symptoms, such as pain or soreness, as it presses against nearby nerves and muscles. If in the abdomen it can cause abdominal pains commonly mistaken for menstrual cramps, indigestion, or cause constipation.
The first reliably documented report of Psilocybe semilanceata intoxication involved a British family in 1799, who prepared a meal with mushrooms they had picked in London's Green Park. According to the chemist Augustus Everard Brande, the father and his four children experienced typical symptoms associated with ingestion, including pupil dilation, spontaneous laughter and delirium. The identification of the species responsible was made possible by James Sowerby's 1803 book Coloured Figures of English Fungi or Mushrooms, which included a description of the fungus, then known as Agaricus glutinosus (originally described by Moses Ashley Curtis in 1780). According to German mycologist Jochen Gartz, the description of the species is "fully compatible with current knowledge about Psilocybe semilanceata." In the early 1960s, the Swiss scientist Albert Hofmann—known for the synthesis of the psychedelic drug LSD—chemically analyzed P. semilanceata fruit bodies collected in Switzerland and France by the botanist Roger Heim. Using the technique of paper chromatography, Hofmann confirmed the presence of 0.25% (by weight) psilocybin in dried samples. Their 1963 publication was the first report of psilocybin in a European mushroom species; previously, it had been known only in Psilocybe species native to Mexico, Asia and North America. This finding was confirmed in the late 1960s with specimens from Scotland and England, Czechoslovakia (1973), Germany (1977), Norway (1978), and Belgium and Finland (1984).

The sequence is Met-Glu-His-Phe-Pro-Gly-Pro, a seven-residue peptide. The first four residues correspond to the ACTH(4-10) fragment, and Pro-Gly-Pro is added at the C-terminus. N-acetylated versions also appear in the literature and differ in mass.
Semax is a synthetic analog of the ACTH(4-10) fragment rather than a copy of the full hormone. Truncation plus the added tail removes the steroidogenic portion of ACTH. That distinction explains why it is studied for nervous-system effects instead of adrenal stimulation.
It is registered as a prescription product in Russia and has been used there clinically, chiefly by the intranasal route. No approval exists in the United States or the European Union. Availability elsewhere typically falls under research-chemical distribution, with varying purity documentation.
