DIPG is a terminal illness, since it has a 5-year survival rate of <1%. The median overall survival of children diagnosed with DIPG is approximately 9 months. The 1- and 2-year survival rates are approximately 30% and less than 10%, respectively. These statistics make DIPG one of the most devastating pediatric cancers. Although 75–85% of patients show some improvement in their symptoms after radiation therapy, DIPGs almost always begin to grow again (called recurrence, relapse, or progression). Clinical trials have reported that the median time between radiation therapy and progression is 5–8.8 months. Patients whose tumors begin to grow again may be eligible for experimental treatment through clinical trials to try to slow or stop the growth of the tumor. However, clinical trials have not shown any significant benefit from experimental DIPG therapies so far. For DIPGs that progress, they usually grow quickly and affect important parts of the brain. The median time from tumor progression to death is usually very short, between 1 and 4.5 months. During this time, doctors focus on palliative care: controlling symptoms and making the patient as comfortable as possible.

=== Research === A major thread of Bumpus' research is determining how antiviral drugs used to treat HIV-1 are metabolized and how genetic variations in drug-processing enzymes may impact these drugs' efficacy. One antiviral commonly used to treat and prevent HIV is tenofovir (Viread). Tenofovir is a nucleotide analog reverse transcriptase inhibitor that prevents the HIV virus from replicating. Tenofovir is given in an inactive form – it has to be phosphorylated twice to become active, and this phosphorylation is carried out in two steps by separate kinases. Bumpus and her team identified which kinases do this and found that the enzymes responsible varied by cell type, so administration route (e.g. orally or topically) could affect how effectively the drug is processed. They also sequenced the genes of these kinases from different patients and found that some people have genetic variants in the kinases that may affect how effectively the drug is processed. Tenofovir can be given prophylactically, and Bumpus collaborated with researchers from around the world in a study of the use of tenofovir for HIV pre-exposure prophylaxis (PrEP) to prevent HIV infection in heterosexual men and women. Bumpus has also researched nonnucleoside reverse transcriptase inhibitors. Her lab was the first to publish the P450-catalyzed phase 1 and phase 2 metabolic pathways of two nonnucleoside reverse transcriptase inhibitors, rilpivirine and etravirine and was first to characterize the metabolism of the nonnucleoside reverse transcriptase inhibitor dapivirine.

In most cases the proton-motive force is generated by an electron transport chain which acts as a proton pump, using the Gibbs free energy of redox reactions to pump protons (hydrogen ions) out across the membrane, separating the charge across the membrane. In mitochondria, energy released by the electron transport chain is used to move protons from the mitochondrial matrix (N side) to the intermembrane space (P side). Moving the protons out of the mitochondrion creates a lower concentration of positively charged protons inside it, resulting in excess negative charge on the inside of the membrane. The electrical potential gradient is about -170 mV , negative inside (N). These gradients - charge difference and the proton concentration difference both create a combined electrochemical gradient across the membrane, often expressed as the proton-motive force (PMF). In mitochondria, the PMF is almost entirely made up of the electrical component but in chloroplasts the PMF is made up mostly of the pH gradient because the charge of protons H+ is neutralized by the movement of Cl− and other anions. In either case, the PMF needs to be greater than about 460 mV (45 kJ/mol) for the ATP synthase to be able to make ATP.

Tasmanian oak is a native Australian hardwood produced by any of three trees, Eucalyptus regnans, Eucalyptus obliqua or Eucalyptus delegatensis, when it is sourced from the Australian state of Tasmania. Despite the common name "oak", none of the species are in the genus Quercus or the oak family Fagaceae. The hardwood timber is light-coloured, ranging from straw to light reddish brown. It is used in construction, including panelling and flooring, for furniture, and also for reconstituted board and high quality paper. When sourced from Victoria, the wood of Eucalyptus regnans and Eucalyptus delegatensis is called Victorian ash. The species are also widely known by their common names. Eucalyptus obliqua is known as stringybark or messmate, Eucalyptus regnans is known as mountain ash, and the closely related Eucalyptus delegatensis is known as alpine ash or woollybutt.
=== Interviews and promotional press === Vulture published an interview with Clipse in which an album was explicitly confirmed, on June 19, 2024—one day after "The Birds Don't Sing" was previewed at the Louis Vuitton's Men's Spring–Summer Show 2025. Interviewed by Craig Jenkins, the duo reflected on the passing of their parents and how that experience, along with Williams's father dealing with dialysis treatments, influenced the lyrics of "The Birds Don't Sing". Pusha T revealed that Malice came up with the title, inspired by writer and activist Maya Angelou's 1969 autobiography I Know Why the Caged Bird Sings. Other topics included the brothers' father giving Malice his blessing to return to Clipse, the "coke rap" label often assigned to them, and Pusha T being referenced by Kendrick Lamar in his 2024 rap battle with Drake. Additionally, a comeback album was officially announced for the first time, although no title or release date was given. Pusha T stated: "The next year is going to be filled with appearances, touring, and a rap album of the year." Clipse spoke to Njera Perkins of People in an interview published on August 15, 2024. They talked about their then-upcoming performance at the Pepsi Dig In Day Block Party charity event, they joys of performing live in general, and how Williams had been pushing for a reunion.
Regarding the first part of the film, Barry White of Citizen Game commented, "Considering the comparatively paltry resources [...] at [the Purchase Brothers] disposal this short still manages to be better than every video game movie currently in existence." Wagner Au of NewTeeVee contended that the first part "is one of those rare viral videos that seems destined to launch a breakout success", and added "non-gamers are likely to be impressed by its rollicking action and bravura special effects." Au believed its popularity was due, in part, to the fact that the video is adapted from Half-Life 2. Patrick Goss, writing for TechRadar, said the in-game footage from the Half-Life 2 series "blended almost seamlessly into [the] live action footage" in the film. Valve, developers of the Half-Life series, approved of the film and publicized it in an announcement on Steam, saying "we were blown away – the production quality certainly exceeds that of the usual community-made movies we see."
== Biochemical structure == Kex2 was first purified and characterized by Charles Brenner and Robert Fuller in 1992. The Kex2 crystal structure was solved by a group led by Dagmar Ringe, Robert Fuller and Gregory Petsko. That of Furin was determined by a group led by Manual Than and Wolfram Bode. The key features of Kex2 and Furin are a subtilisin-related catalytic domain, a specificity pocket that requires the amino acid amino terminal to the scissile bond to be arginine for rapid acylation, and a P-domain carboxy-terminal to the subtilisin domain, which is required for biosynthesis.

Each freeze-thaw cycle can degrade a fraction of the peptide and promote aggregation. Dividing a stock solution into single-use volumes removes repeated cycling as a variable. It also limits the time a thawed solution spends at room temperature.
A high main-peak percentage says little about identity, counter-ion content, or residual solvents. It also does not distinguish an isomer or a closely eluting analog. Mass confirmation and the accompanying documentation cover those gaps.
Salt form changes mass, solubility, and hygroscopicity, and acetate and trifluoroacetate salts behave differently in solution. Storage guidance still centers on -20 °C for powder and lower temperatures for solutions. The certificate should state which salt is present.
